What is HAEMOLYTIC PLAQUE ASSAY?

A method used to detect and enumerate individual cells secreting antibody in vitro. Sheep red cells treated when necessary so as to bind the antibody are mixed with a cell suspension to be assayed in a thin layer of agarose, and incubated so that they can secrete antibody which diffuses on to the red cells. Complement is then added and cells secreting antibody are revealed by the presence of an area of haemolysis around them.
